OBJECTIVES/GOALS: Cachexia is the involuntary and irreversible loss of muscle and fat and is a major cause of morbidity and mortality in head and neck cancer (HNC). It remains a poorly understood disease diagnosed by weight loss and a confluence of symptoms. We explored the metabolic and inflammatory mechanisms of cachexia symptoms via an multiomics network algorithm. METHODS/STUDY POPULATION: Prior to chemoradiotherapy, HNC subjects completed questionnaires and donated blood for untargeted (metabolites) and targeted (lipids and cytokines) assays. Metabolites and lipids were measured by liquid chromatography mass spectrometry. Cytokines were measured by multiplex assays. We plotted a multiomics network graph by estimating partial least squares correlations amongst metabolites, lipids, cytokines, and common cachexia symptoms—max percent weight loss over 1 year, baseline BMI, fatigue, performance, albumin, hemoglobin, and white blood cell count. To interpret the network, an algorithm identified highly correlated clusters of metabolites-lipids-cytokines-symptoms representing possible biological relatedness, which were functionally annotated via metabolic enrichment analysis. RESULTS/ANTICIPATED RESULTS: In 123 subjects (59 years of age, 72% male, 84% white, avg weight loss of 13%), we analyzed 186 metabolites, 54 lipids, 7 cytokines and 7 cachexia symptoms. We required a correlation >0.25 and P-value <.05 to be included in the network graph, resulting in 323 connections and 3 identified clusters. Max weight loss and baseline BMI were in a cluster enriched by unsaturated fatty acid biosynthesis (P<.0001) and arachidonic acid (P=.01) metabolic pathways but not linked to inflammation cytokines. The five other cachexia symptoms were in a cluster with 4 cytokines (C-reactive protein, interleukin 6, IL10, IL1, Tumor necrosis factor receptor 2) and enriched by aminoacyl tRNA (P<.01) and valine biosynthesis (P=.02). We observed no meaningful differences when we stratified the analysis by human papillomavirus. DISCUSSION/SIGNIFICANCE: Cachexia symptoms in head and neck cancer may be linked to specific metabolic dysregulation—weight loss and BMI were linked to fatty acids; fatigue, anemia and others were linked to amino acids and inflammation. This information may allow for the recognition of a cachexic-metabolic subtype or provide novel targets for metabolic intervention.

Observations of radiocarbon (14C) in Earth’s atmosphere and other carbon reservoirs are important to quantify exchanges of CO2 between reservoirs. The amount of 14C is commonly reported in the so-called Delta notation, i.e., Δ14C, the decay- and fractionation-corrected departure of the ratio of 14C to total C from that ratio in an absolute international standard; this Delta notation permits direct comparison of 14C/C ratios in the several reservoirs. However, as Δ14C of atmospheric CO2, Δ14CO2 is based on the ratio of 14CO2 to total atmospheric CO2, its value can and does change not just because of change in the amount of atmospheric14CO2 but also because of change in the amount of total atmospheric CO2, complicating ascription of change in Δ14CO2 to change in one or the other quantity. Here we suggest that presentation of atmospheric 14CO2 amount as mole fraction relative to dry air (moles of 14CO2 per moles of dry air in Earth’s atmosphere), or as moles or molecules of 14CO2 in Earth’s atmosphere, all readily calculated from Δ14CO2 and the amount of atmospheric CO2 (with slight dependence on δ13CO2), complements presentation only as Δ14CO2, and can provide valuable insight into the evolving budget and distribution of atmospheric 14CO2.
Actin-mediated endocytosis in the E-YSL helps drive epiboly in zebrafish

In zebrafish, a punctate band of F-actin is reported to develop in the external yolk syncytial layer (E-YSL) during the latter part of epiboly in zebrafish embryos. Here, electron microscopy (EM) and fluorescence confocal microscopy were conducted to investigate dynamic changes in the E-YSL membrane during epiboly. Using scanning EM, we report that the surface of the E-YSL is highly convoluted, consisting of a complex interwoven network of branching membrane surface microvilli-like protrusions. The region of membrane surface protrusions was relatively wide at 30% epiboly but narrowed as epiboly progressed. This narrowing was coincident with the formation of the punctate actin band. We also demonstrated using immunogold transmission EM that actin clusters were localized at the membrane surface mainly within the protrusions as well as in deeper locations of the E-YSL. Furthermore, during the latter part of epiboly, the punctate actin band was coincident with a region of highly dynamic endocytosis. Treatment with cytochalasin B led to the disruption of the punctate actin band and the membrane surface protrusions, as well as the attenuation of endocytosis. Together, our data suggest that, in the E-YSL, the region encompassing the membrane surface protrusions and its associated punctate actin band are likely to be an integral part of the localized endocytosis, which is important for the progression of epiboly in zebrafish embryos.
Safety and efficacy of KarXT (Xanomeline Trospium) in Schizophrenia in the Phase 3, Randomized, Double-Blind, Placebo-Controlled EMERGENT-2 Trial

Introduction
KarXT combines the M1/M4 preferring muscarinic receptor agonist xanomeline and the peripherally restricted anticholinergic trospium. In the phase 2 EMERGENT-1 study, KarXT met the primary endpoint of a significant reduction in Positive and Negative Syndrome Scale (PANSS) total score through week 5 vs placebo, improved other key secondary efficacy measures, and was generally well tolerated.
MethodsEMERGENT-2 was a phase 3, randomized, double-blind, placebo-controlled, 5-week trial of KarXT in acutely psychotic patients with schizophrenia in the inpatient setting. Eligible patients were randomized 1:1 to KarXT or matched placebo. Dosing of KarXT (mg xanomeline/mg trospium) started at 50 mg/20 mg BID and increased to a maximum of 125 mg/30 mg BID. The primary efficacy endpoint was change from baseline to week 5 in PANSS total score. Key secondary endpoints included change from baseline to week 5 in PANSS positive subscale, PANSS negative subscale, and PANSS negative Marder factor scores compared with placebo. Efficacy analyses were performed using the modified intent-to-treat population (patients with ≥1 dose of study medication, a baseline PANSS assessment, and ≥1 postbaseline PANSS assessment). All patients receiving ≥1 dose of study drug were included in safety analyses.
Results252 US patients were enrolled. KarXT demonstrated a statistically significant and clinically meaningful 9.6-point reduction from baseline to week 5 (effect size=0.61) in PANSS total score vs placebo (p<0.0001); a significant improvement in PANSS total score was demonstrated starting at week 2 (first postbaseline rating) and continued through the study end. KarXT also met key secondary endpoints. Results at week 5 included a 2.9-point reduction in PANSS positive subscale score with KarXT vs placebo (p<0.0001), a 1.8-point reduction in PANSS negative subscale score with KarXT vs placebo (p=0.0055), and a 2.2-point reduction in PANSS negative Marder factor score with KarXT vs placebo (p=0.0022). KarXT was generally well tolerated. Overall discontinuation rates were similar with KarXT (25%) and placebo (21%). The overall treatment-emergent adverse events (TEAEs) rate for KarXT and placebo was 75% and 58%, respectively. Discontinuation rates related to TEAEs were similar between KarXT (7%) and placebo (6%). Rates of serious TEAEs were similar with KarXT and placebo (2%, each group); no serious TEAEs were determined to be drug related. The most common TEAEs (≥5%) with KarXT were all mild to moderate in severity and included constipation, dyspepsia, nausea, vomiting, headache, blood pressure increases, dizziness, gastroesophageal reflux disease, abdominal discomfort, and diarrhea. KarXT was not associated with sedation/somnolence, weight gain, and extrapyramidal symptoms.
ConclusionsKarXT has the potential to be the first in a new class of treatments for patients with schizophrenia and a promising alternative to postsynaptic dopamine D2 receptor antagonists.

Infants and children born with CHD are at significant risk for neurodevelopmental delays and abnormalities. Individualised developmental care is widely recognised as best practice to support early neurodevelopment for medically fragile infants born premature or requiring surgical intervention after birth. However, wide variability in clinical practice is consistently demonstrated in units caring for infants with CHD. The Cardiac Newborn Neuroprotective Network, a Special Interest Group of the Cardiac Neurodevelopmental Outcome Collaborative, formed a working group of experts to create an evidence-based developmental care pathway to guide clinical practice in hospital settings caring for infants with CHD. The clinical pathway, “Developmental Care Pathway for Hospitalized Infants with Congenital Heart Disease,” includes recommendations for standardised developmental assessment, parent mental health screening, and the implementation of a daily developmental care bundle, which incorporates individualised assessments and interventions tailored to meet the needs of this unique infant population and their families. Hospitals caring for infants with CHD are encouraged to adopt this developmental care pathway and track metrics and outcomes using a quality improvement framework.

Understanding place-based contributors to health requires geographically and culturally diverse study populations, but sharing location data is a significant challenge to multisite studies. Here, we describe a standardized and reproducible method to perform geospatial analyses for multisite studies. Using census tract-level information, we created software for geocoding and geospatial data linkage that was distributed to a consortium of birth cohorts located throughout the USA. Individual sites performed geospatial linkages and returned tract-level information for 8810 children to a central site for analyses. Our generalizable approach demonstrates the feasibility of geospatial analyses across study sites to promote collaborative translational research.

In 2017, transgender woman Danica Roem stunned political observers in Virginia by unseating a long-time anti-LGBTQ legislator from a conservative district in the Virginia House of Delegates.1 She was the first openly transgender person elected and seated to a state legislature. Delegate Roem’s election was historic in LGBTQ political representation, but it also occurred in a period when backlash against the LGBTQ community seemed to be growing (Taylor, Lewis, and Haider-Markel 2018). These two threads led us to ask: How are LGBTQ candidates achieving historic successes even as forces seem mobilized against them?

Background:
Sulphur mustard (HD) is a lipophilic caustic alkylating vesicant (blister agent) that has mutagenic and carcinogenic effects. Among the studied perturbations are long-term genitourinary (GU) and fertility effects. Approximately 50,000 Iranian soldiers and civilians were exposed to HD during the Iraq-Iran war (1980-1989). This study questioned the wives of Iraq-Iran war veterans to determine the effects of male HD-exposure on pregnancy complications, adverse pregnancy outcomes, and secondary infertility.
Methods:A retrospective, survey-based cohort study was conducted of wives of Iranian military veterans that survived HD-associated injuries while serving in Ahvaz, Iran during the Iraq-Iran war (1980-1989), as compared to non-exposed veterans serving concomitantly. Patients were identified from a database of injured veterans maintained by the Foundation of Martyrs and Veterans Affairs (Iran) via a systematic random sampling method utilizing a random number table. Using a validated questionnaire, collected data included: demographics; type and severity of chemical injury; spouse’s obstetric history (pregnancy number, duration, complications, and outcomes before and after spouse’s chemical injury); and secondary infertility.
Results:An increase in spontaneous abortion (P = .03), congenital anomalies (P < .0001), and secondary infertility (P = .003) were observed. These findings were greatest amongst those with HD injuries affecting >50% body surface area. No difference in stillbirth, premature birth, or low birth weight was observed.
Conclusion:Exposure to HD in combat may have long-lasting fertility effects on soldiers and their spouses, including spontaneous abortion, congenital anomalies, and secondary infertility. Further investigation is needed into the long-term effects of HD exposure as well as methods to better protect soldiers.

A national need is to prepare for and respond to accidental or intentional disasters categorized as chemical, biological, radiological, nuclear, or explosive (CBRNE). These incidents require specific subject-matter expertise, yet have commonalities. We identify 7 core elements comprising CBRNE science that require integration for effective preparedness planning and public health and medical response and recovery. These core elements are (1) basic and clinical sciences, (2) modeling and systems management, (3) planning, (4) response and incident management, (5) recovery and resilience, (6) lessons learned, and (7) continuous improvement. A key feature is the ability of relevant subject matter experts to integrate information into response operations. We propose the CBRNE medical operations science support expert as a professional who (1) understands that CBRNE incidents require an integrated systems approach, (2) understands the key functions and contributions of CBRNE science practitioners, (3) helps direct strategic and tactical CBRNE planning and responses through first-hand experience, and (4) provides advice to senior decision-makers managing response activities. Recognition of both CBRNE science as a distinct competency and the establishment of the CBRNE medical operations science support expert informs the public of the enormous progress made, broadcasts opportunities for new talent, and enhances the sophistication and analytic expertise of senior managers planning for and responding to CBRNE incidents.
3399 Systematically Integrating Microbiomes and Exposomes for Translational Research

OBJECTIVES/SPECIFIC AIMS: Characterize microbiome metadata describing specimens collected, genomic pipelines and microbiome results, and incorporate them into a data integration platform for enabling harmonization, integration and assimilation of microbial genomics with exposures as spatiotemporal events. METHODS/STUDY POPULATION: We followed similar methods utilized in previous efforts in charactering and developing metadata models for describing microbiome metadata. Due to the heterogeneity in microbiome and exposome data, we aligned them along a conceptual representation of different data used in translational research; microbiomes being biospecimen-derived, and exposomes being a combination of sensor measurements, surveys and computationally modelled data. We performed a review of literature describing microbiome data, metadata, and semantics [4–15], along with existing datasets [16] and developed an initial metadata model. We reviewed the model with microbiome domain experts for its accuracy and completeness, and with translational researchers for its utility in different studies, and iteratively refined it. We then incorporated the logical model into OpenFurther’s metadata repository MDR [17,18] for harmonization of different microbiome datasets, as well as integration and assimilation of microbiome-exposome events utilizing the UPIE. RESULTS/ANTICIPATED RESULTS: Our model for describing the microbiome currently includes three domains (1) the specimen collected for analysis, (2) the microbial genomics pipelines, and (3) details of the microbiome genomics. For (1), we utilized biospecimen data model that harmonizes the data structures of caTissue, OpenSpecimen and other commonly available specimen management platform. (3) includes details about the organisms, isolate, host specifics, sequencing methodology, genomic sequences and annotations, microbiome phenotype, genomic data and storage, genomic copies and associated times stamps. We then incorporated this logical model into the MDR as assets and associations that UPIE utilizes to harmonize different microbiome datasets, followed by integration and assimilation of microbiome-exposome events. Details of (2) are ongoing. DISCUSSION/SIGNIFICANCE OF IMPACT: The role of the microbiome and co-influences from environmental exposures in etio-pathology of various pulmonary conditions isn’t well understood [19–24]. This metadata model for the microbiome provides a systematic approach for integrating microbial genomics with sensor-based environmental and physiological data, and clinical data that are present in varying spatial and temporal granularities and require complex methods for integration, assimilation and analysis. Incorporation of this microbiome model will advance the performance of sensor-based exposure studies of the (UPIE) to support novel research paradigms that will improve our understanding of the role of microbiome in promoting and preventing airway inflammation by performing a range of hypothesis-driven microbiome-exposome pediatric asthma studies across the translational spectrum.

Good education requires student experiences that deliver lessons about practice as well as theory and that encourage students to work for the public good—especially in the operation of democratic institutions (Dewey 1923; Dewy 1938). We report on an evaluation of the pedagogical value of a research project involving 23 colleges and universities across the country. Faculty trained and supervised students who observed polling places in the 2016 General Election. Our findings indicate that this was a valuable learning experience in both the short and long terms. Students found their experiences to be valuable and reported learning generally and specifically related to course material. Postelection, they also felt more knowledgeable about election science topics, voting behavior, and research methods. Students reported interest in participating in similar research in the future, would recommend other students to do so, and expressed interest in more learning and research about the topics central to their experience. Our results suggest that participants appreciated the importance of elections and their study. Collectively, the participating students are engaged and efficacious—essential qualities of citizens in a democracy.
MSMA Antagonizes Glyphosate and Glufosinate Efficacy on Broadleaf and Grass Weeds

Field and greenhouse studies were conducted to investigate the compatibility of MSMA in a tank mixture with glyphosate or glufosinate for broadleaf and grass weed control. Glyphosate, glufosinate, and MSMA were evaluated at 0.5×, 1×, and 2× rates, with 1× rates of 0.84 kgae/ha, 0.5 kgai/ha, and 2.2 kgai/ha, respectively. Glyphosate and glufosinate provided similar levels of control for most weed species and were often more efficacious than MSMA alone. Glyphosate controlled Palmer amaranth better than glufosinate. Glufosinate controlled hemp sesbania, pitted morningglory, and ivyleaf morningglory better than glyphosate at one location. Weed control was not improved with the addition of MSMA to glyphosate or glufosinate when compared with either herbicide alone. MSMA antagonized glyphosate efficacy on barnyardgrass, browntop millet, hemp sesbania, Palmer amaranth, and redroot pigweed. MSMA antagonized glufosinate efficacy on browntop millet, hemp sesbania, ivyleaf morningglory, johnsongrass, Palmer amaranth, pitted morningglory, prickly sida, redroot pigweed, and velvetleaf. Antagonism of glyphosate or glufosinate by MSMA was often overcome by applying the 2× rate of either herbicide alone. MSMA is not a compatible tank-mixture partner with glyphosate or glufosinate for weed control in cotton.

Redstem filaree is a troublesome weed for sugarbeet growers in northern Wyoming and southern Montana. Field studies were conducted in Powell, WY, in 2006 and 2008 to determine the influence of season-long interference of various redstem filaree densities and the duration of interference on sugarbeet. Root and sucrose yield decreased with increasing redstem filaree density. The rectangular hyperbola model with the asymptote (A) bounded at 100% maximum yield reduction characterized the relationship between redstem filaree density and sugarbeet yield reduction. The estimated parameter I (percent yield reduction per unit weed density as density approaches zero) was 5% for root and sucrose yield reduction. Sugarbeet root yield decreased as the duration of redstem filaree interference increased. The critical timing of redstem filaree removal to avoid 5 and 10% root yield reduction was estimated to be 25 and 32 d after sugarbeet emergence, respectively. Redstem filaree interference did not affect the sucrose content percentage. These results demonstrate that redstem filaree is competitive with sugarbeet and should be managed appropriately to reduce negative effects on yield.
Cabbage (Brassica oleracea) Response to Pendimethalin Applied Posttransplant

Field and greenhouse studies were conducted from 1997 to 2001 to determine cabbage response to posttransplant applications of pendimethalin (0.56 to 2.24 kg ai/ha). Differential variety response was minimal, and applications greater than 0.56 kg caused severe and persistent crop injury and reduced head number and yield in ‘Azan’, ‘Storage 4’, ‘Super Elite’, and ‘Super Red 90’. Pendimethalin (1.7 kg) applied posttransplant reduced cabbage yield weights 23, 30, and 87% with bare root, large, and small transplants, respectively. Application (0.84 kg) to soil, foliage, or soil and foliage caused 0, 81, and 82% dry weight reduction by 21 d after treatment, respectively. Anatomical analysis of two-leaf seedlings collected 3 wk after pendimethalin treatment (1.12 kg ai/ha) showed stunting of the shoot apical meristem and its emerging leaves, disorganization of apical structure with disruption of normal cell division and cell expansion, and abnormal differentiation of the vasculature in leaves and hypocotyls.
Wild Buckwheat (Polygonum convolvulus) Interference in Sugarbeet

Field studies were conducted in Powell, WY in 2006 and 2007 to determine the influence of season-long interference of various wild buckwheat densities and duration of interference on sugarbeet. Percent sucrose content was not affected by wild buckwheat interference. Root and sucrose yield loss per hectare increased as wild buckwheat density increased. The estimated percent yield loss as wild buckwheat density approaches infinity was 64 and 61% for root and sucrose yield loss, respectively. The estimated percent yield loss per unit weed density at low weed densities was 6% for both root and sucrose yield loss. Greater durations of wild buckwheat interference had a negative effect on sugarbeet root yield. The critical timing of weed removal (CTWR) to avoid 5 and 10% root yield loss was 32 and 48 d after sugarbeet emergence (DAE), respectively. These results show that wild buckwheat is competitive with sugarbeet and should be managed appropriately to forestall any negative effects on sugarbeet root and sucrose yield.

The rectangular hyperbola model with the asymptote (A) constrained to 100% maximum yield loss characterized the relationship between lanceleaf sage density and sugarbeet yield loss. The estimated parameter I (yield loss per unit weed density as density approaches zero) was 3% for both root and sucrose yield loss. Increasing duration of lanceleaf sage interference had a negative effect on sugarbeet root yield. The critical timing of weed removal to avoid 5 and 10% root yield loss was 37 and 52 d after sugarbeet emergence, respectively. Lanceleaf sage interference did not affect percentage of sucrose content. These results indicate that lanceleaf sage is not as competitive as other weeds but that appropriate control measures should be undertaken to minimize sugarbeet yield loss from interference.

Multiple means of overcoming interspecific competition between transplanted cabbage and interseeded cover crops were studied in field trials conducted from 1995 to 2001. Cover crop species and time of seeding (1995 and 1996), use of supplemental nitrogen (1997 and 1998), and herbicide regulation (1999 and 2001) were evaluated with the objective of integrating soil-improving cover crops into cabbage production while facilitating weed suppression with minimal use of herbicides. Cabbage was cultivated at 10, 10 + 20, or 10 + 20 + 30 d after transplanting (DAT) with or without cover crops (hairy vetch, lana vetch, or oats) sown at the time of the last cultivation. Early interseeding (10 DAT) of all species significantly reduced cabbage yields. Both vetches could be sown 20 or 30 DAT without a yield penalty. However, weed suppression was not consistently greater than cultivation without cover crops. Spring oats were unacceptably competitive, even when sown 30 DAT in some years. With additional nitrogen, cabbage yields were consistently increased, but the increases were not directly related to decreased competition from either weeds or cover crops. The potential for herbicide regulation of cover crops to prevent cabbage yield losses could not be evaluated because cabbage yields were not reduced by cover crops in 1999 and 2001. Although interseeded crops did not generally provide significant in-season weed suppression compared with cultivation alone, the lack of yield penalty and the potential soil-improving qualities of legumes may justify interseeding hairy vetch at 20 DAT in an integrated system.

Sucrose concentration was not affected by Venice mallow interference. The effect of Venice mallow density on sugarbeet root and sucrose yield loss was described by the rectangular hyperbola model. Root and sucrose yield loss increased as Venice mallow density increased. The estimated asymptote, A (percent yield loss as density approaches infinity) was 61% for both root and sucrose yield loss, and the estimated parameter, I (percent yield loss per unit weed density as density approaches zero) was 6% for both root and sucrose yield loss. Sugarbeet root yield decreased as the duration of Venice mallow interference increased. The critical timing of weed removal to avoid 5 and 10% root yield loss was 30 and 43 d after sugarbeet emergence, respectively. Results show that Venice mallow is competitive with sugarbeet implying that it should be managed appropriately to reduce negative effects on yield and prevent seed bank replenishment and re-infestation in subsequent years.

Approximately half of the variation in wellbeing measures overlaps with variation in personality traits. Studies of non-human primate pedigrees and human twins suggest that this is due to common genetic influences. We tested whether personality polygenic scores for the NEO Five-Factor Inventory (NEO-FFI) domains and for item response theory (IRT) derived extraversion and neuroticism scores predict variance in wellbeing measures. Polygenic scores were based on published genome-wide association (GWA) results in over 17,000 individuals for the NEO-FFI and in over 63,000 for the IRT extraversion and neuroticism traits. The NEO-FFI polygenic scores were used to predict life satisfaction in 7 cohorts, positive affect in 12 cohorts, and general wellbeing in 1 cohort (maximal N = 46,508). Meta-analysis of these results showed no significant association between NEO-FFI personality polygenic scores and the wellbeing measures. IRT extraversion and neuroticism polygenic scores were used to predict life satisfaction and positive affect in almost 37,000 individuals from UK Biobank. Significant positive associations (effect sizes <0.05%) were observed between the extraversion polygenic score and wellbeing measures, and a negative association was observed between the polygenic neuroticism score and life satisfaction. Furthermore, using GWA data, genetic correlations of -0.49 and -0.55 were estimated between neuroticism with life satisfaction and positive affect, respectively. The moderate genetic correlation between neuroticism and wellbeing is in line with twin research showing that genetic influences on wellbeing are also shared with other independent personality domains.
